A new method for double bond characterization in lipids by ultraviolet photodissociation mass spectrometry

Lipids are structurally diverse biomolecules with vital functions in biological systems. Some of these functions are closely associated with the specific location of the carbon-carbon double bond in the lipid acyl chains. The chain length and the number of unsaturated carbon-carbon bonds can be determined using conventional MS/MS-based structural elucidation methods employing low or higher energy collision-induced dissociation. However, these dissociation techniques do not provide more subtle structural details, for example, the position of unsaturated bonds. Another type of activation method – ultraviolet photodissociation can be used for this purpose. In this study, a new method for characterization of double bond location in lipids acyl chain was developed using 193 nm ultraviolet photodissociation implemented on Orbitrap Fusion Lumos mass spectrometer. This approach is based on the derivatization of the double bond with bis-(5-iodo-[2]pyridyl)-disulfide, subsequent specific cleavages provide a unique diagnostic pair bearing information about the double bond position.
Localization of double bond positions in triacylglycerols using HPLC/APCI-MS2

Triacylglycerols are among the most abudant classes of lipid. Their chemical, physical and biological characteristics depend on degree of unsaturation and positions of double bonds in acyls. This thesis was focused on localization of double bonds using tandem mass spectrometry with atmospheric pressure chemical ionization. We studied 14 standards and 44 standard mixtures with different number of double bonds and lenght of hydrocarbob chain in acyls. Standards were obtained from commercial sources and prepared of randomizations reaction in microscale. Localization of double bonds was deduced from fragments of molecular aduct with C3H5N+· ([M+55]+·) created in presence of acetonitrile in the mobile phase. This metod was applied for HPLC/MS analysis of natural mixtures of triacylglycerols. Aldrithiol-2 as a new derivatization agent for double bond localization in unsaturated compounds by mass spectrometry

A new method for localizing double bonds in lipids using derivatization with 2,2'-clipyridyl disulfide (Aldrithio-2) and electrospray ionization mass spectrometry (EST-MS) is presented. The derivatization introduces a permanent charge to the molecules. When collisionally activated, the derivatives provide diagnostic peaks useful for establishing the position of the original double bond. Since permanent charge of the derivative significantly increases the detectability of analytes, the method works well also for of very low polarity like hydrocarbons.

Art Therapy - Working with Standard Population

The topic will be divided into two parts {--} theoretical and practical ones {--} and a conclusion with an evaluation of the project. The theoretical part will include the basic data concerning developmental psychology together with a deeper insight into a period of adulthood and middle- and post-middle ages. It will be an outlook of a healthy population that deals with common situations of everyday life, with a period of their and their children´s maturity, with a choice of their first partner for life, with a departure of the partner and finally with their children´s leaving and arranging their own lives. My work will include some systematic procedures and casuistics among the selected clients. I will focuse on solving the common situations of everyday life in a Simulation studio together with an effort to approach a so called {\clqq}Healthy Rožnov Drawing``. The practical part will analyse the client´s work. In a conclusion I want to summarise my long-term activities with the above mentioned clients.
